Abstract: Mammals are colonized by an astronomical number of commensal microorganisms on their environmental exposed surfaces. These symbiotic species build up a complex community that aids their hosts in several physiological activities. We have shown that lack of intestinal microbiota is accompanied by a state of active IL-10–mediated inflammatory hyporesponsiveness. The present study investigated whether the germfree state and its hyporesponsive phenotype alter host resistance to an infectious bacterial insult. Experiments performed in germfree mice infected with Klebsiella pneumoniae showed that these animals are drastically susceptible to bacterial infection in an IL-10–dependent manner. In germfree mice, IL-10 restrains proinflammatory mediator production and neutrophil recruitment and favors pathogen growth and dissemination. Germfree mice were resistant to LPS treatment. However, priming of these animals with several TLR agonists recovered their inflammatory responsiveness to sterile injury. LPS pretreatment also rendered germfree mice resistant to pulmonary K. pneumoniae infection, abrogated IL-10 production, and restored TNF-α and CXCL1 production and neutrophil mobilization into lungs of infected germfree mice. This effective inflammatory response mounted by LPS-treated germfree mice resulted in bacterial clearance and enhanced survival upon infection. Therefore, host colonization by indigenous microbiota alters the way the host reacts to environmental infectious stimuli, probably through activation of TLR-dependent pathways. Symbiotic gut colonization enables proper inflammatory response to harmful insults to the host, and increases resilience of the entire mammal-microbiota consortium to environmental pressures.

Abstract: An analytical representation for first and subsequent stroke currents is proposed, and simple application examples are given. The waveforms are able to reproduce the characteristic concave profile at the front of first stroke currents, including or not a second peak that is usually observed in the data available from direct measurements on actual lightning. They also reproduce, with good accuracy, median parameters describing lightning currents measured at short instrumented towers. The proposed waveforms have the advantage of using a sum of Heidler functions that last over the entire time of analysis. This avoids the use of separate functions to characterize the front and the tail of the synthesized currents and makes the proposed waveforms suitable for lightning computational analysis.

Abstract: Non-profits, as well as the media, have hypothesized the existence of a radicalization pipeline on YouTube, claiming that users systematically progress towards more extreme content on the platform. Yet, there is to date no substantial quantitative evidence of this alleged pipeline. To close this gap, we conduct a large-scale audit of user radicalization on YouTube. We analyze 330,925 videos posted on 349 channels, which we broadly classified into four types: Media, the Alt-lite, the Intellectual Dark Web (I.D.W.), and the Alt-right. According to the aforementioned radicalization hypothesis, channels in the I.D.W. and the Alt-lite serve as gateways to fringe far-right ideology, here represented by Alt-right channels. Processing 72M+ comments, we show that the three channel types indeed increasingly share the same user base; that users consistently migrate from milder to more extreme content; and that a large percentage of users who consume Alt-right content now consumed Alt-lite and I.D.W. content in the past. We also probe YouTube's recommendation algorithm, looking at more than 2M video and channel recommendations between May/July 2019. We find that Alt-lite content is easily reachable from I.D.W. channels, while Alt-right videos are reachable only through channel recommendations. Overall, we paint a comprehensive picture of user radicalization on YouTube.
