Detailed Mechanistic Insights into HIV-1 Sensitivity to Three Generations of Fusion Inhibitors

TL;DR: This work combined virological, computational, and biophysical experiments with comprehensive mutational analyses and tested resistance to T20 and second and third generation inhibitors (T1249 and T2635), finding that most amino acid substitutions caused resistance to the first generation peptide T20.
Journal ArticleDOI

Structure-function relationships in an antifreeze polypeptide. The role of neutral, polar amino acids.

TL;DR: It is concluded that a specific arrangement of both threonine and asparagine (or aspartic acid) residues is critical for maximal activity and that the AFPs probably bind to the pyramidal faces of ice with a specific orientation.
Journal ArticleDOI

Structural and functional analysis of interhelical interactions in the human immunodeficiency virus type 1 gp41 envelope glycoprotein by alanine-scanning mutagenesis.

TL;DR: Results suggest that the Leu-565 and Val-570 residues are important determinants of conserved packing interactions between the amino- and carboxyl-terminal helices of gp41, and propose that the high degree of sequence conservation at Gly-572 and Arg-579 may result from selective pressures imposed by prefusogenic conformations of the HIV-1 envelope glycoprotein.
Journal ArticleDOI

Functional and structural characterization of recombinant dermcidin-1L, a human antimicrobial peptide.

TL;DR: Assays demonstrated that dermcidin-1L displayed microbicidal activity against several opportunistic nosocomial pathogens, but no hemolytic activity against human erythrocytes even at concentrations up to 100 microM, andStructural studies performed by circular dichroism spectroscopy indicated that the secondary structure of dermcodin- 1L was very flexible, and both alpha-helix and beta-sheet structures might be required for the antimicrobial activity.
Journal ArticleDOI

Secondary structures of a new class of lipid body proteins from oilseeds.

TL;DR: A model of the general structure of oleosins is proposed whereby the central hydrophobic domain of the protein with a predominantly beta-strand structure is embedded into the non-aqueous phase of lipid-bodies.
