Abstract: In normal rats and mice, immunostaining with specific antibodies revealed that nuclei of most prostatic epithelial cells harbor estrogen receptor beta (ERbeta). In rat ventral prostate, 530- and 549-aa isoforms of the receptor were identified. These sediment in the 4S region of low-salt sucrose gradients, indicating that prostatic ERbeta does not contain the same protein chaperones that are associated with ERalpha. Estradiol (E(2)) binding and ERbeta immunoreactivity coincide on the gradient, with no indication of ERalpha. In prostates from mice in which the ERbeta gene has been inactivated (BERKO), androgen receptor (AR) levels are elevated, and the tissue contains multiple hyperplastic foci. Most epithelial cells express the proliferation antigen Ki-67. In contrast, prostatic epithelium from wild-type littermates is single layered with no hyperplasia, and very few cells express Ki-67. Rat ventral prostate contains an estrogenic component, which comigrates on HPLC with the testosterone metabolite 5alpha-androstane-3beta,17beta-diol (3betaAdiol). This compound, which competes with E(2) for binding to ERbeta and elicits an estrogenic response in the aorta but not in the pituitary, decreases the AR content in prostates of wild-type mice but does not affect the elevated levels seen in ERbeta knockout (BERKO) mice. Thus ERbeta, probably as a complex with 3betaAdiol, is involved in regulating the AR content of the rodent prostate and in restraining epithelial growth. These findings suggest that ligands specific for ERbeta may be useful in the prevention and/or clinical management of prostatic hyperplasia and neoplasia.

Abstract: Tropical forests include a diversity of habitats, which has led to specialization in plants. Near Iquitos, in the Peruvian Amazon, nutrient-rich clay forests surround nutrient-poor white-sand forests, each harboring a unique composition of habitat specialist trees. We tested the hypothesis that the combination of impoverished soils and herbivory creates strong natural selection for plant defenses in white-sand forest, while rapid growth is favored in clay forests. Recently, we reported evidence from a reciprocal-transplant experiment that manipulated the presence of herbivores and involved 20 species from six genera, including phylogenetically independent pairs of closely related white-sand and clay specialists. When protected from herbivores, clay specialists exhibited faster growth rates than white-sand specialists in both habitats. But, when unprotected, white-sand specialists outperformed clay specialists in white- sand habitat, and clay specialists outperformed white-sand specialists in clay habitat. Here we test further the hypothesis that the growth-defense trade-off contributes to habitat specialization by comparing patterns of growth, herbivory, and defensive traits in these same six genera of white-sand and clay specialists. While the probability of herbivore attack did not differ between the two habitats, an artificial defoliation experiment showed that the impact of herbivory on plant mortality was significantly greater in white-sand forests. We quantified the amount of terpenes, phenolics, leaf toughness, and available foliar protein for the plants in the experiment. Different genera invested in different defensive strategies, and we found strong evidence for phylogenetic constraint in defense type. Overall, however, we found significantly higher total defense investment for white-sand specialists, relative to their clay specialist congeners. Furthermore, herbivore resistance consistently exhibited a significant trade-off against growth rate in each of the six phylogenetically independent species-pairs. These results confirm theoretical predictions that a trade-off exists between growth rate and defense investment, causing white-sand and clay specialists to evolve divergent strategies. We propose that the growth-defense trade-off is universal and provides an important mechanism by which herbivores govern plant distribution patterns across resource gradients.

Abstract: We analyze 40 cosmological re-simulations of individual massive galaxies with present-day stellar masses of M∗ > 6.3 × 10 10 M⊙ in order to investigate the physical origin of the observed strong increase in galaxy sizes and the decrease of the stellar velocity dispersions since redshift z ≈ 2. At present 25 out of 40 galaxies are quiescent with structural parameters (sizes and velocity dispersions) in agreement with local early type galaxies. At z=2 all simulated galaxies with M∗ & 10 11 M⊙ (11 out of 40) at z=2 are compact with projected half-mass radii of ≈ 0.77 (±0.24) kpc and line-of-sight velocity dispersions within the projected half-mass radius of ≈ 262 (±28) kms −1 (3 out of 11 are already quiescent). Similar to observed compact early-type galaxies at high redshift the simulated galaxies are clearly offset from the local mass-size and mass-velocity dispersion relations. Towards redshift zero the sizes increase by a factor of ∼ 5 − 6, following R1/2 ∝ (1 + z) α with α = −1.44 for quiescent galaxies (α = −1.12 for all galaxies). The velocity dispersions drop by about one-third since z ≈ 2 , following σ1/2 ∝ (1 + z) β with β = 0.44 for the quiescent galaxies (β = 0.37 for all galaxies). The simulated size and dispersion evolution is in good agreement with observations and results from the subsequent accretion and merging of stellar systems at z . 2 which is a natural consequence of the hierarchical structure formation. A significant number of the simulated massive galaxies (7 out of 40) experience no merger more massive than 1:4 (usually considered as major mergers). On average, the dominant accretion mode is stellar minor mergers with a mass-weighted mass-ratio of 1:5. We therefore conclude that the evolution of massive early-type galaxies since z ≈ 2 and their present-day properties are predominantly determined by frequent ’minor’ mergers of moderate mass-ratios and not by major mergers alone.

Abstract: The global loss of biodiversity continues at an alarming rate. Genomic approaches have been suggested as a promising tool for conservation practice as scaling up to genome-wide data can improve traditional conservation genetic inferences and provide qualitatively novel insights. However, the generation of genomic data and subsequent analyses and interpretations remain challenging and largely confined to academic research in ecology and evolution. This generates a gap between basic research and applicable solutions for conservation managers faced with multifaceted problems. Before the real-world conservation potential of genomic research can be realized, we suggest that current infrastructures need to be modified, methods must mature, analytical pipelines need to be developed, and successful case studies must be disseminated to practitioners.

Abstract: Background— Endothelial vasodilator dysfunction and carotid intima-media thickening (IMT) are 2 indicators of subclinical cardiovascular disease. We examined their correlation and interaction with risk factors in a large, community-based cohort of young adults. Methods and Results— As part of the longitudinal Cardiovascular Risk in Young Finns Study, we measured endothelium-dependent brachial artery flow–mediated dilatation (FMD) and carotid artery IMT by ultrasound in 2109 healthy adults aged 24 to 39 years. FMD was inversely associated with IMT (P≤0.001) in a multivariate model adjusted for age, sex, brachial vessel size, and several risk variables. The subjects with age- and sex-specific FMD values in the extreme deciles were classified into groups of impaired (n=204, FMD=1.1±1.4%, mean±SD) and enhanced (n=204, FMD=16.3±2.9%) FMD response.
