Mapping Glutathione Utilization in the Developing Zebrafish (Danio Rerio) Embryo

TL;DR: Monochlorobimane (MCB), a dye conjugated with GSH by glutathione-s-transferase (GST) to form a fluorescent adduct, was used to visualize organ-specific GSH utilization in live developing zebrafish embryos and can be applied to identify sensitive target tissues of toxicants.

The role of arachidonic acid in the regulation of nitric oxide synthase isoforms by HIV gp120 protein in astroglial cells.

TL;DR: It is demonstrated that HIV-1 gp120 is able to inhibit neuronal NOS through a cytosolic phospholipase A2 (cPLA2)-dependent arachidonic acid (AA) production, this response being critical for allowing activation of the transcriptional factor NF-κB and subsequent iNOS and interleukin-1β transcription in astroglial cells.
Journal ArticleDOI

Prooxidant and antioxidant properties of salicylaldehyde isonicotinoyl hydrazone iron chelators in HepG2 cells.

TL;DR: Activation by SIH iron chelators of a hormetic antioxidant response contributes to their antioxidant properties and modulates the anti- and pro-oxidant balance.
Journal ArticleDOI

The role of TLR4/NF-κB signaling in the radioprotective effects of exogenous Prdx6.

TL;DR: In this paper, the authors investigated the role of TLR4/NF-κB signaling pathway in the radioprotective effect of recombinant peroxiredoxin 6 (Prdx6) and its mutant form, Prdx6-C47S, which lacks peroxidase activity.
Journal ArticleDOI

Spectrophotometric versus spectrofluorometric assessment in the study of the relationships between lipid peroxidation and metabolic dysregulation

TL;DR: The results show that these optimized protocols are suitable for complex in vivo studies, as they require low quantities of sample and reagents, and are sensitive, rapid, and economical, with the option of adapting them for high‐throughput analysis.

ROS as signalling molecules: mechanisms that generate specificity in ROS homeostasis

TL;DR: The pathways that regulate ROS homeostasis are crucial for mitigating the toxicity of ROS and provide strong evidence about specificity in ROS signalling.

Evaluation of the probe 2',7'-dichlorofluorescin as an indicator of reactive oxygen species formation and oxidative stress.

TL;DR: Data suggest that H2O2-Fe(2+)-derived oxidant is mainly responsible for the nonenzymatic oxidation of DCFH, which remains an attractive probe as an overall index of oxidative stress in toxicological phenomena.
Journal ArticleDOI

Reconciling the chemistry and biology of reactive oxygen species

TL;DR: This review examines how target selectivity and antioxidant effectiveness vary for different oxidants and highlights areas where greater understanding is required on the fate of oxidants generated by cellular NADPH oxidases and on the identification of oxidant sensors in cell signaling.

Flow cytometric studies of oxidative product formation by neutrophils: a graded response to membrane stimulation.

TL;DR: The data indicate that the DCFH oxidation assay is quantitatively related to the oxidative metabolic burst of PMNL, and they strongly suggest that the reaction is mediated by H2O2 generated by the PMNL.
