Measuring reactive oxygen and nitrogen species with fluorescent probes: challenges and limitations.

TLDR
A critical analysis of the challenges and limitations of the most widely used fluorescent probes for detecting and measuring reactive oxygen and nitrogen species and proposed guidelines that will help present and future researchers with regard to the optimal use of selected fluorescent probes and interpretation of results are presented.

Journal ArticleDOI

γ-Glutamyltransferase enzyme activity of cancer cells modulates L-γ-glutamyl-p-nitroanilide (GPNA) cytotoxicity.

TL;DR: The GGT-catalyzed hydrolysis of GPNA produced cytotoxic effects in lung cancer A549 cells, resulting from the release of metabolite p-nitroaniline (PNA) rather than from the inhibition of Gln uptake, indicating the mechanism of action of GP NA is more complex than believed.

Phthalocyanine-based photosensitizer with tumor-pH-responsive properties for cancer theranostics

TL;DR: A pH-responsive photosensitizer synthesized by conjugating zinc phthalocyanine with 2,4,6-tris(N,N-dimethylaminomethyl) phenoxy (TAP), which exhibited high phototoxicity at pH 6.5 but no obvious photot toxicity at physiological pH 7.4, may open up a new avenue for tumor-targeted and image-guided cancer theranostics in PDT.

Fluorescence microscopy as a tool for visualization of metal-induced oxidative stress in plants

TL;DR: In this paper, the authors compared five ROS-related and two non-ROS-related reagents in the roots of Zea mays germinated and growing in 1 or 100µM cadmium-enriched solution.
Journal ArticleDOI

Redox biology in pulmonary arterial hypertension (2013 Grover Conference Series).

TL;DR: This review has attempted to summarize some of the important existing studies, to point out areas of overlap between oxidative stress and metabolic dysfunction, and to do so under the unifying heading of redox biology, to advance the discussion of carbon-substrate metabolism beyond a focus on glucose and its fate in the cell to encompass other carbon substrates.
Journal ArticleDOI

Myostatin knockout induces apoptosis in human cervical cancer cells via elevated reactive oxygen species generation.

TL;DR: Limiting the production and function of Mstn may be an effective therapeutic intervention during cancer progression and muscle loss in cachexia because of its role in regulating mitochondrial metabolism and apoptosis within cancer cells.

ROS as signalling molecules: mechanisms that generate specificity in ROS homeostasis

TL;DR: The pathways that regulate ROS homeostasis are crucial for mitigating the toxicity of ROS and provide strong evidence about specificity in ROS signalling.

Evaluation of the probe 2',7'-dichlorofluorescin as an indicator of reactive oxygen species formation and oxidative stress.

TL;DR: Data suggest that H2O2-Fe(2+)-derived oxidant is mainly responsible for the nonenzymatic oxidation of DCFH, which remains an attractive probe as an overall index of oxidative stress in toxicological phenomena.
Journal ArticleDOI

Reconciling the chemistry and biology of reactive oxygen species

TL;DR: This review examines how target selectivity and antioxidant effectiveness vary for different oxidants and highlights areas where greater understanding is required on the fate of oxidants generated by cellular NADPH oxidases and on the identification of oxidant sensors in cell signaling.

Flow cytometric studies of oxidative product formation by neutrophils: a graded response to membrane stimulation.

TL;DR: The data indicate that the DCFH oxidation assay is quantitatively related to the oxidative metabolic burst of PMNL, and they strongly suggest that the reaction is mediated by H2O2 generated by the PMNL.
