Circular RNAs Are the Predominant Transcript Isoform from Hundreds of Human Genes in Diverse Cell Types

TLDR
By deep sequencing of RNA from a variety of normal and malignant human cells, this work suggests that a non-canonical mode of RNA splicing, resulting in a circular RNA isoform, is a general feature of the gene expression program in human cells.
Abstract
Most human pre-mRNAs are spliced into linear molecules that retain the exon order defined by the genomic sequence. By deep sequencing of RNA from a variety of normal and malignant human cells, we found RNA transcripts from many human genes in which the exons were arranged in a non-canonical order. Statistical estimates and biochemical assays provided strong evidence that a substantial fraction of the spliced transcripts from hundreds of genes are circular RNAs. Our results suggest that a non-canonical mode of RNA splicing, resulting in a circular RNA isoform, is a general feature of the gene expression program in human cells.

Profiles and Bioinformatics Analysis of Differentially Expressed Circrnas in Taxol-Resistant Non-Small Cell Lung Cancer Cells

TL;DR: This study evaluated the roles of circRNAs in the chemoresistance of non-small cell lung cancer (NSCLC) to taxol and found that circRNas exert biological effects by absorbing and sequestering miRNA molecules.

CircRNA_0000392 promotes colorectal cancer progression through the miR-193a-5p/PIK3R3/AKT axis.

TL;DR: Functional studies revealed that reducing the expression of circRNA_0000392 could inhibit the proliferation and invasion of CRC both in vitro and in vivo, suggesting that circRNA-0000392 is a potential therapeutic target for the treatment of colorectal cancer and a predictive marker for CRC patients.

Cyclic RNA hsa‑circ‑000595 regulates apoptosis of aortic smooth muscle cells

TL;DR: In this paper, aortic aneurysm is a cardiovascular condition with a serious risk of mortality and the dismal prognosis of any type of major cardiovascular disease and the results of the present study laid an epigenetic foundation for exploring the underlying mechanisms of the development of aortia, and it was determined that miR-19a is a target of hsa-circ-000595.

Expression dynamics, relationships, and transcriptional regulations of diverse transcripts in mouse spermatogenic cells

TL;DR: The identified 20,639 message RNAs, 7,168 long non-coding RNAs and 15,101 circular RNAs in mouse spermatogenic cells, and found many of them were specifically expressed in testes, and showed that the productions of circRNAs and piRNAs were highly regulated instead of random processes.

A coding-independent function of gene and pseudogene mRNAs regulates tumour biology

TL;DR: It is found that PTENP1 is biologically active as it can regulate cellular levels of PTEN and exert a growth-suppressive role, and this analysis extended to other cancer-related genes that possess pseudogenes, and revealed a non-coding function for mRNAs.
