TLDR
Escherichia coli cells are 4--6 times more transformable and 20--30 times more competent after 24 h incubation in cold calcium chloride than immediately after calcium chloride treatment.

Expression of invertase activity in Yarrowia lipolytica and its use as a selective marker.

TL;DR: A gene fusion where the Saccharomyces cerevisiae SUC2 gene is placed under the control of the promoter and signal sequence of the Y. lipolytica XPR2 gene, which encodes an Alkaline Extracellular Protease (AEP) and it was shown that this chimeric gene could be used as a dominant marker for transformation in a one-step procedure.

Hormonal regulation of plasminogen activator mRNA production in porcine kidney cells.

TL;DR: M measuring PA-mRNA sequences in the Xenopus oocyte translation system showed a 15-20-fold enhanced PA-synthesizing capacity when supplied with poly(A)+RNA from induced cells, above that obtained from uninduced cell RNA, suggesting that increased concentrations of cAMP mediate the inducing effects of the hormones by enhancing the transcription of PA- mRNA sequences.

Transformation of Streptococcus mutans with chromosomal and shuttle plasmid (pYA629) DNAs.

TL;DR: Two incompatibility groups among S. mutans strains that are readily transformable, display maximal cariogenicity in gnotobiotic rats, and give high scores for in vitro measures of important virulence attributes have been identified to facilitate studies on the genetic basis and control of virulence.
Journal ArticleDOI

Intramolecular inhibition of human defensin HNP-1 by its propiece.

TL;DR: Addition of unlinked propiece inhibited the bactericidal and cell-permeabilizing activity of mature HNP-1 in a dose-dependent manner, and acts as an efficient intramolecular inhibitor of defensin H NP-1 cytotoxicity.

5′ flanking sequence signals are required for activity of silkworm alanine tRNA genes in homologous in vitro transcription systems

TL;DR: It is found that at least two regions of DNA are required for accurate transcription of tRNA2Ala genes by homologous B. mori extracts, and one is retained in the truncated gene and may be within the tRNA coding sequence.

Construction and characterization of new cloning vehicles. II. A multipurpose cloning system.

TL;DR: In vitro recombination techniques were used to construct a new cloning vehicle, pBR322, which is a relaxed replicating plasmid, does not produce and is sensitive to colicin E1, and carries resistance genes to the antibiotics ampicillin (Ap) and tetracycline (Tc).

Nonchromosomal Antibiotic Resistance in Bacteria: Genetic Transformation of Escherichia coli by R-Factor DNA

TL;DR: Covalently-closed, catenated, and open (nicked) circular forms of R-factor DNA are all effective in transformation, but denaturation and sonication abolish the transforming ability of R.factor DNA in this system.
Journal ArticleDOI

Calcium-dependent bacteriophage DNA infection.

TL;DR: Escherichia coli cells of strain K12 and C can be made competent to take up temperate phage DNA without the use of “helper phage”, and is effective for both linear and circular DNA molecules.
