TLDR
Recent advances have shown that in two MAP kinase pathways (the mating response pathway in the fission yeast Schizosaccharomyces pombe, and receptor tyrosine kinase signalling), the small GTP binding protein ras p21 links membrane events to kinase pathway activation.

Global, in vivo, and site-specific phosphorylation dynamics in signaling networks.

TL;DR: A general mass spectrometric technology is developed and applied for identification and quantitation of phosphorylation sites as a function of stimulus, time, and subcellular location to provide a missing link in a global, integrative view of cellular regulation.
Journal ArticleDOI

A protein kinase involved in the regulation of inflammatory cytokine biosynthesis.

TL;DR: Production of interleukin-1 and tumour necrosis factor from stimulated human monocytes is inhibited by a new series of pyridinyl-imidazole compounds, suggesting that the CSBPs are critical for cytokine production.

Protein modules and signalling networks

TL;DR: This work highlights conserved protein domains that act as key regulatory participants in many of these different signalling pathways in multicellular organisms.
Journal ArticleDOI

Mammalian thioredoxin is a direct inhibitor of apoptosis signal-regulating kinase (ASK) 1.

TL;DR: Evidence that Trx is a negative regulator of ASK1 suggests possible mechanisms for redox regulation of the apoptosis signal transduction pathway as well as the effects of antioxidants against cytokine‐ and stress‐induced apoptosis.

Activation of the c-Raf protein kinase by protein kinase C phosphorylation.

TL;DR: Re recombinant baculoviruses expressing PKC and Raf polypeptides are shown to be able to activate c-Raf in a TPA-dependent manner upon coexpression in insect cells, suggesting that c- Raf acts immediately downstream of PKC in a protein kinase cascade which is triggered by TPA and may lead to transcriptional activation of T PA-inducible genes and tumor promotion.

Constitutive mutants of the protein kinase STE11 activate the yeast pheromone response pathway in the absence of the G protein.

TL;DR: From a ste4 deletion strain, both dominant and recessive suppressors that show increased transcription of pheromone responsive genes and have regained the ability to mate, albeit at a low level are isolated.
Journal ArticleDOI

Reconstitution of the Raf-1-MEK-ERK signal transduction pathway in vitro.

TL;DR: It is concluded that MEK is a direct substrate of Raf-1 and that the activation of MEK by Raf- 1 is due to phosphorylation by Raf -1, which is sufficient for MEK activation.
Journal ArticleDOI

MAP kinase activator from insulin-stimulated skeletal muscle is a protein threonine/tyrosine kinase

TL;DR: MAP kinase kinase is the first established example of a protein kinase that can phosphorylate an exogenous protein on threonine as well as tyrosine residues.
