TLDR
A research strategy to achieve the connection matrix of the human brain (the human “connectome”) is proposed, and its potential impact is discussed.Abstract:
The connection matrix of the human brain (the human “connectome”) represents an indispensable foundation for basic and applied neurobiological research. However, the network of anatomical connections linking the neuronal elements of the human brain is still largely unknown. While some databases or collations of large-scale anatomical connection patterns exist for other mammalian species, there is currently no connection matrix of the human brain, nor is there a coordinated research effort to collect, archive, and disseminate this important information. We propose a research strategy to achieve this goal, and discuss its potential impact.read more

TL;DR: A brain-wide, cellular-level, mesoscale connectome for the mouse, using enhanced green fluorescent protein-expressing adeno-associated viral vectors to trace axonal projections from defined regions and cell types, and high-throughput serial two-photon tomography to image the EGFP-labelled axons throughout the brain.

Neurodegenerative Diseases Target Large-Scale Human Brain Networks
TL;DR: It is shown that five different neurodegenerative syndromes cause circumscribed atrophy within five distinct, healthy, human intrinsic functional connectivity networks, and a direct link between intrinsic connectivity and gray matter structure is discovered.

Large-scale brain networks in cognition: emerging methods and principles
Steven L. Bressler,Vinod Menon +1 more
TL;DR: It is argued that the emerging science of large-scale brain networks provides a coherent framework for understanding of cognition that allows a principled exploration of how cognitive functions emerge from, and are constrained by, core structural and functional networks of the brain.

TL;DR: It is shown that CLARITY enables fine structural analysis of clinical samples, including non-sectioned human tissue from a neuropsychiatric-disease setting, establishing a path for the transmutation of human tissue into a stable, intact and accessible form suitable for probing structural and molecular underpinnings of physiological function and disease.

Neurophysiological and Computational Principles of Cortical Rhythms in Cognition
TL;DR: A plethora of studies will be reviewed on the involvement of long-distance neuronal coherence in cognitive functions such as multisensory integration, working memory, and selective attention, and implications of abnormal neural synchronization are discussed as they relate to mental disorders like schizophrenia and autism.

Dynamic causal modelling.
TL;DR: As with previous analyses of effective connectivity, the focus is on experimentally induced changes in coupling, but unlike previous approaches in neuroimaging, the causal model ascribes responses to designed deterministic inputs, as opposed to treating inputs as unknown and stochastic.
