Abstract: Repeated administration of cocaine to rodents produces a progressive augmentation in motor activity known as behavioral sensitization. By using microdialysis in the ventral striatum, some studies have found that the development of behavioral sensitization is associated with a similar augmentation in dopamine release, while others have not. It was postulated that differences in doses and withdrawal periods may account for the discrepancies between studies. Rats were behaviorally sensitized to daily peripheral injections using two cocaine treatment regimens (15 mg/kg, i.p. x 5 d or 30 mg/kg, i.p. x 5 d). Using in vivo microdialysis in the ventral striatum, the effect of acute cocaine (15 mg/kg, i.p.) on extracellular dopamine content and motor behavior was examined at various times after discontinuing daily treatments. Twenty-four hours after discontinuing the low dose of daily cocaine, the increase in motor activity and extracellular dopamine elicited by an acute cocaine challenge was significantly elevated. In contrast, following the higher daily treatment regimen there was a significant augmentation in motor activity, but the increase in extracellular dopamine produced by cocaine was significantly reduced. When rats were challenged 10-14 d after discontinuing either dosage regimen of daily cocaine, the increase in both motor activity and extracellular dopamine was augmented. In general, the increase in extracellular dopamine by an acute cocaine challenge increased over time when rats were challenged between 1 and 22 d after discontinuing daily cocaine. Basal concentrations of extracellular dopamine were determined by measuring the in vivo flux of dopamine across the dialysis membrane, and there was no significant difference at 24 hr or 2 weeks following the last daily injection of saline or cocaine. It is concluded that behavioral sensitization to cocaine is generally associated with an augmentation in extracellular dopamine in the ventral striatum, but that high doses of daily cocaine produce apparent tolerance to the augmentation in extracellular dopamine during the early withdrawal period.

Abstract: Innate immunity is initiated in animals and plants through the recognition of a variety of pathogen-associated molecules that in animals are called pathogen-associated molecular patterns and in plants are called elicitors. Some plant pathogen-derived elicitors have been identified as peptides, but peptide elicitors derived from the plant itself that activate defensive genes against pathogens have not been previously identified. Here, we report the isolation and characterization of a 23-aa peptide from Arabidopsis, called AtPep1, which activates transcription of the defensive gene defensin (PDF1.2) and activates the synthesis of H2O2, both being components of the innate immune response. The peptide is derived from a 92-aa precursor encoded within a small gene that is inducible by wounding, methyl jasmonate, and ethylene. Constitutive expression of the AtPep1 precursor gene PROPEP1 in transgenic Arabidopsis plants causes a constitutive transcription of PDF1.2. When grown in soil, the transgenic plants exhibited an increased root development compared with WT plants and an enhanced resistance toward the root pathogen Pythium irregulare. Six paralogs of PROPEP1 are present in Arabidopsis, and orthologs have been identified in species of several agriculturally important plant families, where they are of interest for their possible use in crop improvement.

Abstract: Fresh food products can be processed using UV light as a germicidal medium to reduce the food-borne microbial load. Water has been treated with UV light to obtain drinking water for quite some time. Pumpable fruit and vegetable products are generally very suitable for processing by UV light to reduce the microbial load. Today, most of these products are pasteurised to obtain microbiologically safe and nutritious products. However, pasteurisation can change the taste and flavour of such products because of the temperature and processing time. Juices from different sources can be treated by exposure to UV light at different doses. On the other hand, variables such as flow rate, exposure time, type of fruit product, juice colour and juice composition, among other variables, need to be studied to obtain fruit products with reduced microbial load, increased shelf life and adequate sensory and nutritional characteristics. Reduction of microbial load through UV light application as a disinfection medium for food products other than liquids is also being studied. Moreover, UV technology could be a source for pasteurisation of liquids, or disinfection of solid foods as an alternative technology, instead of thermal treatment or application of antimicrobial compounds.

Abstract: The last glacial period was terminated by an abrupt warming event in the North Atlantic ∼15,000 years before the present, and warming events of similar age have been reported from low latitudes. Understanding the mechanism of this termination requires that the precise relative timing of abrupt climate warming in the tropics versus the North Atlantic be known. Nitrogen and argon isotopes in trapped air in Greenland ice show that the Greenland Summit warmed 9 ± 3°C over a period of several decades, beginning 14,672 years ago. Atmospheric methane concentrations rose abruptly over a ∼50-year period and began their increase 20 to 30 years after the onset of the abrupt Greenland warming. These data suggest that tropical climate became warmer or wetter (or both) ∼20 to 80 years after the onset of Greenland warming, supporting a North Atlantic rather than a tropical trigger for the climate event.
