Cell division is required for de novo methylation of CpG islands in bladder cancer cells.
Mihaela Velicescu,Daniel J. Weisenberger,Felicidad A. Gonzales,Yvonne C. Tsai,Carvell T. Nguyen,Peter A. Jones +5 more
TL;DR: It is suggested that cell division is required for de novo methylation of CpG islands and that DNMT3a may play a role in methylating CpGs poor regions or repetitive DNA elements outside of the S phase of the cell cycle.
Journal ArticleDOI
Evidence for the role of promoter methylation in the regulation of MMP-9 gene expression.
Éric Chicoine,Pierre-Olivier Estève,Olivier Robledo,Céline Van Themsche,Edouard F. Potworowski,Yves St-Pierre +5 more
TL;DR: The results indicate that methylation may contribute to the transcriptional activity of the MMP-9 promoter and confirm the fact that DNA methylation exerts suppression on transcriptionalActivity.
Journal ArticleDOI
The CpG island promoter of the human proopiomelanocortin gene is methylated in nonexpressing normal tissue and tumors and represses expression.
TL;DR: It is demonstrated that methylation in vitro is sufficient for silencing of expression, which is not reversed by treatment with Trichostatin A, suggesting that inhibition of expression may be mediated by means other than recruitment of histone deacetylase activity.
Journal ArticleDOI
Alterations of DNA methylation by glutathione depletion
TL;DR: The results provide strong evidence that GSH-depleting agents significantly impair cytosine methylation and indicate that alterations in gene expression could result from a high dose and/or prolonged exposure to G SH-depletting agents, e.g. medications, chemotherapeutic agents and environmental toxins.
Journal ArticleDOI
Dual Function of Drosophila Cells as APCs for Naive CD8+ T Cells: Implications for Tumor Immunotherapy
Siquan Sun,Zeling Cai,Pierre Langlade-Demoyen,Hiroshi Kosaka,Anders Brunmark,Michael R. Jackson,Per A. Peterson,Jonathan Sprent +7 more
TL;DR: It is shown that Ld-transfected Drosophila cells are able to induce autologous spleen cells to respond to a tumor-specific peptide in vitro and, after transfer, cause tumor rejection in vivo.

Journal ArticleDOI
Targeted mutation of the DNA methyltransferase gene results in embryonic lethality.
TL;DR: Results indicate that while a 3-fold reduction in levels of genomic m5C has no detectable effect on the viability or proliferation of ES cells in culture, a similar reduction of DNA methylation in embryos causes abnormal development and embryonic lethality.
Journal ArticleDOI
CpG-rich islands and the function of DNA methylation
TL;DR: It is likely that most vertebrate genes are associated with ‘HTF islands’—DNA sequences in which CpG is abundant and non-methylated; however, highly tissue-specific genes, though, usually lack islands.
Journal ArticleDOI
Purification, sequence, and cellular localization of a novel chromosomal protein that binds to Methylated DNA
Joe D. Lewis,Joe D. Lewis,Richard R. Meehan,Richard R. Meehan,William J. Henzel,Ingrid Maurer-Fogy,Peter Jeppesen,Franz Klein,Adrian Bird,Adrian Bird +9 more
TL;DR: This work reports the identification, purification, and cDNA cloning of a novel MeCP called MeCP2, which unlike MeCP1, the new protein is able to bind to DNA that contains a single methyl-CpG pair.
Journal ArticleDOI
Multiple new phenotypes induced in 10T1/2 and 3T3 cells treated with 5-azacytidine.
Shirley M. Taylor,Peter A. Jones +1 more
TL;DR: Three new mesenchymal phenotypes were expressed in cultures of Swiss 3T3 and C3H/10T1/2CL8 mouse cells treated with 5-azacytidine or 5-aza-2'-deoxycytidine, implying that cell division was obligatory for the expression of the new phenotypes.
Journal ArticleDOI
DNA methylation inhibits transcription indirectly via a methyl-CpG binding protein
Joan Boyes,Adrian Bird +1 more
TL;DR: Repression of transcription in vitro for four different promoters was shown to be an indirect effect and the mediator of repression had properties indistinguishable from those of a methyl-CpG binding protein (MeCP-1) that has been previously identified.
