Abstract: Efficient genome editing with Cas9-sgRNA in vivo has required the use of viral delivery systems, which have limitations for clinical applications. Translational efforts to develop other RNA therapeutics have shown that judicious chemical modification of RNAs can improve therapeutic efficacy by reducing susceptibility to nuclease degradation. Guided by the structure of the Cas9-sgRNA complex, we identify regions of sgRNA that can be modified while maintaining or enhancing genome-editing activity, and we develop an optimal set of chemical modifications for in vivo applications. Using lipid nanoparticle formulations of these enhanced sgRNAs (e-sgRNA) and mRNA encoding Cas9, we show that a single intravenous injection into mice induces >80% editing of Pcsk9 in the liver. Serum Pcsk9 is reduced to undetectable levels, and cholesterol levels are significantly lowered about 35% to 40% in animals. This strategy may enable non-viral, Cas9-based genome editing in the liver in clinical settings.

Abstract: The effects of light and nitrogen deficiency on biomass, fatty acid content and composition were studied in Parietochloris incisa, the unicellular freshwater chlorophyte accumulating very high amounts of arachidonic-acid-rich triacylglycerols. P. incisa cultures grown on complete nutrient medium and under high light (400 μmol photons m− 2 s−1) showed the highest rate of growth in comparison to medium (200 μmol photons m−2 s−1) and low (35 μmol photons m−2 s−1) light intensity. Cultures grown under high light (on complete BG-11 medium) attained higher volumetric contents of total fatty acids and arachidonic acid due to greater increase in biomass. Nitrogen starvation brought about a strong increase in the arachidonic acid proportion of total fatty acids. Thus, adjustments to cultivation conditions could serve as an efficient tool for manipulation of yield and relative content of arachidonic acid in P. incisa. The significance of the changes in lipid metabolism for adaptation of P. incisa to high-light stress and nitrogen deficiency is also discussed.

Abstract: The Interstellar Boundary Explorer (IBEX) is a small explorer mission that launched on 19 October 2008 with the sole, focused science objective to discover the global interaction between the solar wind and the interstellar medium. IBEX is designed to achieve this objective by answering four fundamental science questions: (1) What is the global strength and structure of the termination shock, (2) How are energetic protons accelerated at the termination shock, (3) What are the global properties of the solar wind flow beyond the termination shock and in the heliotail, and (4) How does the interstellar flow interact with the heliosphere beyond the heliopause? The answers to these questions rely on energy-resolved images of energetic neutral atoms (ENAs), which originate beyond the termination shock, in the inner heliosheath. To make these exploratory ENA observations IBEX carries two ultra-high sensitivity ENA cameras on a simple spinning spacecraft. IBEX’s very high apogee Earth orbit was achieved using a new and significantly enhanced method for launching small satellites; this orbit allows viewing of the outer heliosphere from beyond the Earth’s relatively bright magnetospheric ENA emissions. The combination of full-sky imaging and energy spectral measurements of ENAs over the range from ∼10 eV to 6 keV provides the critical information to allow us to achieve our science objective and understand this global interaction for the first time. The IBEX mission was developed to provide the first global views of the Sun’s interstellar boundaries, unveiling the physics of the heliosphere’s interstellar interaction, providing a deeper understanding of the heliosphere and thereby astrospheres throughout the galaxy, and creating the opportunity to make even greater unanticipated discoveries.

Abstract: This paper describes a measurement of the Z/gamma* boson transverse momentum spectrum using ATLAS proton-proton collision data at a centre-of-mass energy of root s = 7TeV at the LHC. The measurement is performed in the Z/gamma* -> e(+)e(-) and Z/gamma* -> mu(+)mu(-) channels, using data corresponding to an integrated luminosity of 4.7 fb(-1). Normalized differential cross sections as a function of the Z/gamma* boson transverse momentum are measured for transverse momenta up to 800 GeV. The measurement is performed inclusively for Z/gamma* rapidities up to 2.4, as well as in three rapidity bins. The channel results are combined, compared to perturbative and resummed QCD calculations and used to constrain the parton shower parameters of Monte Carlo generators.
