Owen W. Griffith

- Vol. 27, Iss: 9, pp 922-935
TLDR
Because GSH plays a critical role in cellular defenses against electrophiles, oxidative stress and nitrosating species, pharmacologic manipulation of GSH synthesis has received much attention.

Excess Dietary l-Cysteine, but Not l-Cystine, Is Lethal for Chicks but Not for Rats or Pigs

TL;DR: The results lend credence to the acute toxic effects associated with the ingestion of excess sulfur amino acids and highlight the potential for excess dietary cyst(e)ine to be more pernicious than Met in certain species.
Journal ArticleDOI

Structure Prediction and Active Site Analysis of the Metal Binding Determinants in γ-Glutamylcysteine Synthetase

TL;DR: Homology is detected between γ-GCS and glutamine synthetase (GS), allowing these proteins to be unified into a superfamily of carboxylate-amine/ammonia ligases, demonstrating that the nature of the metal ligands plays an important role in metal specificity.
Journal ArticleDOI

The gender-dependent difference of liver GSH antioxidant system in mice and its influence on isoline-induced liver injury.

TL;DR: The results demonstrated the higher GCL and GPx activities in female mice, which indicated their crucial roles in regulating the resistance of liver injury induced by hepatotoxins in female, and revealed the female-resistant liver injuryinduced by HPAs for the first time.
Journal ArticleDOI

Elevation of glutathione levels by ammonium ions in primary cultures of rat astrocytes.

TL;DR: It is indicated that pathophysiological concentrations of ammonium ions result in increased astroglial levels of GSH which may affect the metabolism and function of astrocytes.
Journal ArticleDOI

Chronic iron overload enhances inducible nitric oxide synthase expression in rat liver

TL;DR: It is concluded that CIO triggers liver oxidative stress at early times, with upregulation of iNOS expression involving the ERK/NF-kappaB pathway at later times, a finding that may represent a hepatoprotective mechanism against CIO toxicity in addition to the recovery of GSH homeostasis.

Antioxidant and Redox Regulation of Gene Transcription

TL;DR: The efficacy of different antioxidants to favorably influence the molecular mechanisms implicated in human disease should be a critical determinant of its selection for clinical studies.

Efficacy of oral N-acetylcysteine in the treatment of acetaminophen overdose. Analysis of the national multicenter study (1976 to 1985)

TL;DR: It is concluded that N-acetylcysteine treatment should be started within eight hours of an acetaminophen overdose, but that treatment is still indicated at least as late as 24 hours after ingestion, and it may be superior when treatment is delayed.
