Biologic and pharmacologic regulation of mammalian glutathione synthesis.

Owen W. Griffith
- 01 Nov 1999 -
- Vol. 27, Iss: 9, pp 922-935
TLDR
Because GSH plays a critical role in cellular defenses against electrophiles, oxidative stress and nitrosating species, pharmacologic manipulation of GSH synthesis has received much attention.

Cadmium-mediated oxidative stress in alveolar epithelial cells induces the expression of γ-glutamylcysteine synthetase catalytic subunit and glutathione S-transferase α and π isoforms: Potential role of activator protein-1

TL;DR: Cadmium-induced oxidative stress, assessed by alterations in GSH homeostasis and an accelerated rate of intracellular oxidant production, could constitute early events in the signal transduction pathway mediating these responses, and the dimeric transcription factor, activator protein-1, may also play a regulatory role in this process.

Glutathione levels and enzyme activity in the tissues of bank vole Clethrionomys glareolus chronically exposed to a mixture of metal contaminants

TL;DR: Evidence indicates that the most sensitive parameter of metal toxicity for animals living in a chronically contaminated environment is the GSH/GSSG ratio, which was decreased in the liver of animals with high Cd levels.

Cocaine-induced adaptations in cellular redox balance contributes to enduring behavioral plasticity.

TL;DR: Cocaine-induced oxidative stress induces changes in GSTpi that contribute to cocaine-induced behavioral plasticity, indicating that cocaine- induced behavioral neuroplasticity may be a factor in relapse.

Diphenyleneiodonium Triggers the Efflux of Glutathione from Cultured Cells

TL;DR: It is concluded that DPI induces a dramatic efflux of cellular GSH from T24 cells via a specific transport channel, which provides a potential mechanism for its proapoptotic effect, and it also has important implications for the regulation of glutathione homeostasis in cells.
Journal ArticleDOI

Ferroptosis caused by cysteine insufficiency and oxidative insult.

TL;DR: It appears that xCT functions as the gatekeeper for determining cellular survival by regulating the availability of Cys in the cell, and the excessive activation of p53 induces ferroptosis by suppressing the expression of xCT in genetic and nongenetic manners.

Efficacy of oral N-acetylcysteine in the treatment of acetaminophen overdose. Analysis of the national multicenter study (1976 to 1985)

TL;DR: It is concluded that N-acetylcysteine treatment should be started within eight hours of an acetaminophen overdose, but that treatment is still indicated at least as late as 24 hours after ingestion, and it may be superior when treatment is delayed.
