Owen W. Griffith

- Vol. 27, Iss: 9, pp 922-935
TLDR
Because GSH plays a critical role in cellular defenses against electrophiles, oxidative stress and nitrosating species, pharmacologic manipulation of GSH synthesis has received much attention.

Chloroform, carbon tetrachloride and glutathione depletion induce secondary genotoxicity in liver cells via oxidative stress

TL;DR: It is hypothesised that certain substances, at high concentrations, can induce an oxidative stress via the depletion of glutathione (GSH) and other antioxidant defences and that this may lead to indirect genotoxicity, that could contribute to carcinogenicity.
Journal ArticleDOI

Antioxidant defenses in killifish (Fundulus heteroclitus) exposed to contaminated sediments and model prooxidants: short-term and heritable responses.

TL;DR: Larval first- and second-generation offspring of Elizabeth River killifish were more resistant to the toxicity of t-butyl hydroperoxide than were King's Creek offspring, indicating a heritable tolerance of exposure to oxidative stress.
Journal ArticleDOI

Molecular mechanisms of cisplatin resistance in bladder cancer.

TL;DR: These include methods to reduce cisplatin bioavailability within a cell, and defects in the machinery that produces cell death following cisPlatin-induced DNA damage, which could be used to predict response in individual tumors.
Journal ArticleDOI

Redox regulation and its emerging roles in stem cells and stem-like cancer cells.

TL;DR: Evidence that supports the involvement of redox homeostasis for stem cell self-renewal, differentiation, and survival are reviewed and the possibility of exploiting the redox status in cancer stem cells as a novel therapeutic strategy is discussed.

Antioxidant and Redox Regulation of Gene Transcription

TL;DR: The efficacy of different antioxidants to favorably influence the molecular mechanisms implicated in human disease should be a critical determinant of its selection for clinical studies.

Efficacy of oral N-acetylcysteine in the treatment of acetaminophen overdose. Analysis of the national multicenter study (1976 to 1985)

TL;DR: It is concluded that N-acetylcysteine treatment should be started within eight hours of an acetaminophen overdose, but that treatment is still indicated at least as late as 24 hours after ingestion, and it may be superior when treatment is delayed.
