TLDR
The functional requirement of EMT and/or MET during the individual steps of tumor metastasis is reviewed and the potential of targeting this program when treating metastatic diseases is discussed.Abstract:
Tumor metastasis is a multistep process by which tumor cells disseminate from their primary site and form secondary tumors at a distant site. Metastasis occurs through a series of steps: local invasion, intravasation, transport, extravasation, and colonization. A developmental program termed epithelial-mesenchymal transition (EMT) has been shown to play a critical role in promoting metastasis in epithelium-derived carcinoma. Recent experimental and clinical studies have improved our knowledge of this dynamic program and implicated EMT and its reverse program, mesenchymal-epithelial transition (MET), in the metastatic process. Here, we review the functional requirement of EMT and/or MET during the individual steps of tumor metastasis and discuss the potential of targeting this program when treating metastatic diseases.read more

Regulation of breast cancer biomarkers by intrinsic and extrinsic factors
TL;DR: It is found that adipocytes promote a mesenchymal-to-epithelial-like transition in BC cells suggesting that secondary tumour formation might be promoted by adipocytes, which will aid in understanding the intrinsic and extrinsic factors that should be considered in the future development of combination drug treatments to successfully cure BC.

TL;DR: In this article , the authors assessed the efficacy of Pirfenidone (PFD), an FDA-approved medication for the treatment of idiopathic pulmonary fibrosis, on triple negative breast cancer (TNBC) cells as well as its anti-tumour effects in xenograft tumour model.

Spontaneous formation and spatial self-organization of mechanically induced mesenchymal-like cells within geometrically confined cancer cell monolayers
TL;DR: In this article , the authors developed a micropattern-based cell monolayer model to investigate the regulation of mechanics heterogeneity on the cell phenotypic switch and reveal that mechanics heterogeneity itself is enough to spontaneously induce the emergence of mesenchymal-like phenotype and asymmetrical activation of TGF-β-SMAD signaling.

Identification of circulating tumor cells with EML4-ALK translocation using fluorescence in situ hybridization in advanced ALK-positive patients with lung cancer.
TL;DR: Isolation and culturing of circulating tumor cells (CTCs) may be an alternative to tumor biopsies for the diagnosis of ALK rearrangement and may have clinical applications including serial blood sampling for the development of personalized cancer therapy based on individual genomic information.

Role of the p53/miR-34a pathway in the response to tumor hypoxia
TL;DR: The PPP1R11 (protein phosphatase 1 regulatory inhibitor subunit 11) was identified and characterized as a target of MIR34A, and strategies to target this pathway might be developed to inhibit CRC metastasis and overcome resistance to therapy associated with hypoxia.

TL;DR: Processes similar to the EMTs associated with embryo implantation, embryogenesis, and organ development are appropriated and subverted by chronically inflamed tissues and neoplasias and the identification of the signaling pathways that lead to activation of EMT programs during these disease processes is providing new insights into the plasticity of cellular phenotypes.
