The functional requirement of EMT and/or MET during the individual steps of tumor metastasis is reviewed and the potential of targeting this program when treating metastatic diseases is discussed.Abstract:
Tumor metastasis is a multistep process by which tumor cells disseminate from their primary site and form secondary tumors at a distant site. Metastasis occurs through a series of steps: local invasion, intravasation, transport, extravasation, and colonization. A developmental program termed epithelial-mesenchymal transition (EMT) has been shown to play a critical role in promoting metastasis in epithelium-derived carcinoma. Recent experimental and clinical studies have improved our knowledge of this dynamic program and implicated EMT and its reverse program, mesenchymal-epithelial transition (MET), in the metastatic process. Here, we review the functional requirement of EMT and/or MET during the individual steps of tumor metastasis and discuss the potential of targeting this program when treating metastatic diseases.read more

A Feedback loop comprising EGF/TGF-α Sustains TFCP2-mediated Breast Cancer Progression
Yi Zhao,Neha Kaushik,Jae Hyeok Kang,Nagendra Kumar Kaushik,Seung Han Son,Nizam Uddin,Min Jung Kim,Chul Geun Kim,Su Jae Lee +8 more
TL;DR: It is shown that TFCP2 is a potent factor essential for EMT, stemness, and metastasis in breast cancer, and a novel regulatory mechanism in which a positive feedback loop comprising EGF/TGFα and AKT can control malignant breast cancer progression.

Dynamic extracellular matrix stiffening induces a phenotypic transformation and a migratory shift in epithelial cells.
TL;DR: This work demonstrates the key role extracellular matrix stiffening has in tumor progression through integrin signaling and, in particular, its ability to drive EMT-related changes and metastasis.

TL;DR: In this paper, the authors apply archetype analysis and evolutionary theory to single-cell transcriptomics and show that SCLC cells reside within a cell-state continuum rather than in discrete subtype clusters.

TL;DR: Survival analyses on a subcohort of patients with locally advanced breast cancer uniformly treated in a prospective study showed significantly different outcomes for the patients belonging to the various groups, including a poor prognosis for the basal-like subtype and a significant difference in outcome for the two estrogen receptor-positive groups.

The basics of epithelial-mesenchymal transition

TL;DR: Processes similar to the EMTs associated with embryo implantation, embryogenesis, and organ development are appropriated and subverted by chronically inflamed tissues and neoplasias and the identification of the signaling pathways that lead to activation of EMT programs during these disease processes is providing new insights into the plasticity of cellular phenotypes.
