Generation and analysis of interleukin-4 deficient mice.
TLDR
Some but not all of the in vitro properties of IL-4 are critical for the physiology of the immune system in vivo, but the serum levels of IgG1 and IgE are strongly reduced.Abstract:
Interleukin-4 (IL-4) promotes the growth and differentiation of many hematopoietic cells in vitro; in particular, it directs the immunoglobulin (Ig) class switch to IgG1 and IgE. Mice homozygous for a mutation that inactivates the IL-4 gene were generated to test the requirement for IL-4 in vivo. In the mutant mice T and B cell development was normal, but the serum levels of IgG1 and IgE were strongly reduced. The IgG1 dominance in a T cell-dependent immune response was lost, and IgE was not detectable upon nematode infection. Thus, some but not all of the in vitro properties of IL-4 are critical for the physiology of the immune system in vivo.read more

Interleukin-10-deficient mice develop chronic enterocolitis
TL;DR: The results indicate that the bowel inflammation in the mutants originates from uncontrolled immune responses stimulated by enteric antigens and that IL-10 is an essential immunoregulator in the intestinal tract.

Insulin-promoter-factor 1 is required for pancreas development in mice
TL;DR: The findings show that IPF1 is needed for the formation of the pancreas and suggest that it acts to determine the fate of common pancreatic precursor cells and/ or to regulate their propagation.

Stat6 Is Required for Mediating Responses to IL-4 and for the Development of Th2 Cells
TL;DR: It is demonstrated that, despite the existence of multiple signaling pathways activated by IL-4, Stat6 is essential for mediating responses toIL-4 lymphocytes.

Localization of murine Ig-1b and Ig-1a (IgG 2a) allotypic determinants detected with monoclonal antibodies.

Class switch recombination is IgG1 specific on active and inactive IgH loci of IgG1-secreting B-cell blasts.
TL;DR: It is shown here that IgG1-secreting B cells isolated from such cultures have undergone a similar DNA rearrangement of the switch regions (S mu, S gamma 1) of the Ig heavy chain constant region genes C mu and C gamma 1 on both active and inactive IgH loci.

TL;DR: There is a fairly high degree of homology between mouse interleukin 4 and mouse IL-2 genes extending more than 200 bp upstream of a "TATA" like sequence located 20 bp downstream of the transcription initiation site that may play an important role in the regulated expression of this gene in concanavalin A or antigen-stimulated T lymphocytes.
