Generation and analysis of interleukin-4 deficient mice.
TLDR
Some but not all of the in vitro properties of IL-4 are critical for the physiology of the immune system in vivo, but the serum levels of IgG1 and IgE are strongly reduced.Abstract:
Interleukin-4 (IL-4) promotes the growth and differentiation of many hematopoietic cells in vitro; in particular, it directs the immunoglobulin (Ig) class switch to IgG1 and IgE. Mice homozygous for a mutation that inactivates the IL-4 gene were generated to test the requirement for IL-4 in vivo. In the mutant mice T and B cell development was normal, but the serum levels of IgG1 and IgE were strongly reduced. The IgG1 dominance in a T cell-dependent immune response was lost, and IgE was not detectable upon nematode infection. Thus, some but not all of the in vitro properties of IL-4 are critical for the physiology of the immune system in vivo.read more

In vivo biological effects of recombinant soluble interleukin-4 receptor.
Charles R. Maliszewski,Timothy A. Sato,Davison Barry L,Cindy A. Jacobs,Fred D. Finkelman,William C. Fanslow +5 more
TL;DR: Recombinant slL-4R can not only antagonize functions mediated by endogenous IL-4, but also potentiate the biological activity of exogenously administered IL- 4, which may have possible clinical implications for the recombinant molecule, as well as for natural slL,4R immunoregulation.

Synthesis and regulation of trans-mRNA encoding the immunoglobulin epsilon heavy chain
Akira Shimizu,Tasuku Honjo +1 more
TL;DR: The detected immunoglobulin heavy‐chain trans‐mRNA of the ∊ class is detected in which the variable region of the human transgenic μ chain is correctly spliced to the first exon of the endogenous mouse ∊ constant region, consistent with the previous hypothesis that trans-mRNA is synthesized by a trans‐splicing mechanism.

Expression of a Targeted λ1 Light Chain Gene Is Developmentally Regulated and Independent of Igκ Rearrangements

TL;DR: It is concluded that developmentally controlled transcription of a gene rearrangement in the Igλ locus occurs in the presence of an unrearranged Igκ locus and is therefore not negatively regulated by the latter.

Tissue-specific insulator function at H19/Igf2 revealed by deletions at the imprinting control region
TL;DR: It is suggested that insulator activity of the H19/Igf2 ICR varies by cell type and may depend on cell-specific enhancers as well as posttranslational modifications of the insulator protein CTCF.

Daniel P. Bailey,Mohit Kashyap,Paria Mirmonsef,L.Andrew Bouton,Jos Domen,Jingfang Zhu,Emmanuel N. Dessypris,John J. Ryan +7 more
TL;DR: IL-4 exerts potent apoptotic effects on developing mast cells and monocyte/macrophages through mitochondrial damage and Stat6 activation.
