T-cell quality in memory and protection: implications for vaccine design
TLDR
The importance of using multiparameter flow cytometry to better understand the functional capacity of effector and memory T-cell responses, thereby enabling the development of preventative and therapeutic vaccine strategies for infections is highlighted.Abstract:
T cells mediate effector functions through a variety of mechanisms. Recently, multiparameter flow cytometry has allowed a simultaneous assessment of the phenotype and multiple effector functions of single T cells; the delineation of T cells into distinct functional populations defines the quality of the response. New evidence suggests that the quality of T-cell responses is crucial for determining the disease outcome to various infections. This Review highlights the importance of using multiparameter flow cytometry to better understand the functional capacity of effector and memory T-cell responses, thereby enabling the development of preventative and therapeutic vaccine strategies for infections.read more

Differences in the influenza-specific CD4 T cell immunodominance hierarchy and functional potential between children and young adults.
TL;DR: It is found that the influenza protein-specific immunodominance pattern in children differs from that in young adults, with much lower reactivity to the NP internal virion protein in young children.

TL;DR: In this paper, the authors investigated the role of CD4 and CD8 T cells in memory and protection against leishmaniasis infection and found that the clinical improvement of patients is significantly associated with the participation of the T cells involved in the recovery of the immune suppression throughout the VL treatment.

Effector CD8+ T Cells Are Generated in Response to an Immunodominant Epitope in Type III Effector YopE during Primary Yersinia pseudotuberculosis Infection
TL;DR: Following intravenous infection of naïve C57BL/6 mice with a yopE GAP mutant (the R144A mutant), flow cytometry analysis of splenocytes by tetramer and intracellular cytokine staining over a time course showed that YopE69–77-specific CD8+ T cells producing gamma interferon and tumor necrosis factor alpha were generated by day 7, with a peak at day 14.

Poly-functional and long-lasting anticancer immune response elicited by a safe attenuated Pseudomonas aeruginosa vector for antigens delivery
Xavier Chauchet,Dalil Hannani,Sophia Djebali,David Laurin,Benoit Polack,Jacqueline Marvel,Laurent Buffat,Bertrand Toussaint,Audrey Le Gouellec +8 more
TL;DR: It is demonstrated that P. aeruginosa vector is a safe and promising approach for active and specific antitumor immunotherapy, and multiple immunizations allowed the development of a long-lasting immune response, represented by a pool of predominantly effector memory cells which protected mice against late tumor challenge.

Changes in Functional but Not Structural Avidity during Differentiation of CD8+ Effector Cells In Vivo after Virus Infection
TL;DR: The data presented in this study show that at the peak of the effector response generated after poxvirus infection, SLEC were of higher functional avidity than their MPEC counterpart, however, over time, SleC exhibited a decrease in peptide sensitivity.

Two types of murine helper T cell clone. I. Definition according to profiles of lymphokine activities and secreted proteins.
TL;DR: A panel of antigen-specific mouse helper T cell clones was characterized according to patterns of lymphokine activity production, and two types of T cell were distinguished.

Two subsets of memory T lymphocytes with distinct homing potentials and effector functions
TL;DR: It is shown that expression of CCR7, a chemokine receptor that controls homing to secondary lymphoid organs, divides human memory T cells into two functionally distinct subsets, which are named central memory (TCM) and effector memory (TEM).

TL;DR: Recombinant human RANTES, Mip-1α, and MIP-1β induced a dose-dependent inhibition of different strains of HIV-1, HIV-2, and simian immunodeficiency virus (SIV) and may have relevance for the prevention and therapy of AIDS.

TL;DR: Gko mice have been developed which fail to produce IFN-gamma (gko), because of a targeted disruption of the gene for IFNs, and succumb to a rapid and fatal course of tuberculosis that could be delayed, but not prevented, by treatment with exogenous recombinant IFN.

TL;DR: The expression of distinct sets of TLRs and the corresponding difference in reactivity to microbial molecules among subsets of pre-DCs and imDCs support the concept that they have developed through distinct evolutionary pathways to recognize different microbial antigens.
