T-cell quality in memory and protection: implications for vaccine design
TLDR
The importance of using multiparameter flow cytometry to better understand the functional capacity of effector and memory T-cell responses, thereby enabling the development of preventative and therapeutic vaccine strategies for infections is highlighted.Abstract:
T cells mediate effector functions through a variety of mechanisms. Recently, multiparameter flow cytometry has allowed a simultaneous assessment of the phenotype and multiple effector functions of single T cells; the delineation of T cells into distinct functional populations defines the quality of the response. New evidence suggests that the quality of T-cell responses is crucial for determining the disease outcome to various infections. This Review highlights the importance of using multiparameter flow cytometry to better understand the functional capacity of effector and memory T-cell responses, thereby enabling the development of preventative and therapeutic vaccine strategies for infections.read more

Parasite Antigen-Specific Regulation of Th1, Th2, and Th17 Responses in Strongyloides stercoralis Infection
Rajamanickam Anuradha,Saravanan Munisankar,Chandrakumar Dolla,Paul Kumaran,Thomas B. Nutman,Subash Babu +5 more
TL;DR: S. stercoralis infection is characterized by a parasite Ag-dependent regulation of monofunctional and dual-functional Th1, Th2, and Th17 cells, a regulation also reversible by antihelminthic treatment.

TL;DR: A next generation bivalent human adenovirus serotype 5 (hAd5) vaccine capable of inducing immunity in patients with pre-existing adenavirus immunity, comprising both an S sequence optimized for cell surface expression (S-Fusion) and a conserved nucleocapsid (N) antigen designed to be transported to the endosomal subcellular compartment, with the potential to generate durable immune protection.

Priming of Qualitatively Superior Human Effector CD8+ T Cells Using TLR8 Ligand Combined with FLT3 Ligand.
Anna Lissina,Anna Lissina,Olivia Briceño,Olivia Briceño,Georgia Afonso,Georgia Afonso,Georgia Afonso,Martin Larsen,Martin Larsen,Emma Gostick,David Price,Roberto Mallone,Victor Appay,Victor Appay +13 more
TL;DR: An original model of in vitro CD8+ T cell priming is introduced, based on an accelerated dendritic cell coculture system, which uses unfractionated human PBMCs as the starting material and demonstrates that a selective TLR8 agonist, in combination with FLT3L, primes high-quality CD8- T cell responses.

Latency antigen α-crystallin based vaccination imparts a robust protection against TB by modulating the dynamics of pulmonary cytokines.
Bappaditya Dey,Ruchi Jain,Aparna Khera,Umesh D. Gupta,V. M. Katoch,V. D. Ramanathan,Anil K. Tyagi +6 more
TL;DR: This study shows that “rBCG prime - DNA boost” strategy (R/D) confers a markedly superior protection along with reduced pathology in comparison to BCG vaccination in guinea pigs and in mice, and indicates the superiority of α-crystallin based R/D regimen over BCG.

Two subsets of memory T lymphocytes with distinct homing potentials and effector functions
TL;DR: It is shown that expression of CCR7, a chemokine receptor that controls homing to secondary lymphoid organs, divides human memory T cells into two functionally distinct subsets, which are named central memory (TCM) and effector memory (TEM).

TL;DR: Gko mice have been developed which fail to produce IFN-gamma (gko), because of a targeted disruption of the gene for IFNs, and succumb to a rapid and fatal course of tuberculosis that could be delayed, but not prevented, by treatment with exogenous recombinant IFN.

Norimitsu Kadowaki,Stephen Ho,Svetlana Antonenko,Rene de Waal Malefyt,Robert A. Kastelein,J. Fernando Bazan,Yong-Jun Liu +6 more
TL;DR: The expression of distinct sets of TLRs and the corresponding difference in reactivity to microbial molecules among subsets of pre-DCs and imDCs support the concept that they have developed through distinct evolutionary pathways to recognize different microbial antigens.
