TLDR
Novel analytical approaches — in particular, liquid chromatography and mass spectrometry — for systems-level analysis of lipids and their interacting partners (lipidomics) now make this field a promising area of biomedical research, with a variety of applications in drug and biomarker development.
Abstract
The crucial role of lipids in cell, tissue and organ physiology is demonstrated by a large number of genetic studies and by many human diseases that involve the disruption of lipid metabolic enzymes and pathways. Examples of such diseases include cancer, diabetes, as well as neurodegenerative and infectious diseases. So far, the explosion of information in the fields of genomics and proteomics has not been matched by a corresponding advancement of knowledge in the field of lipids, which is largely due to the complexity of lipids and the lack of powerful tools for their analysis. Novel analytical approaches--in particular, liquid chromatography and mass spectrometry--for systems-level analysis of lipids and their interacting partners (lipidomics) now make this field a promising area of biomedical research, with a variety of applications in drug and biomarker development.

Binary Matrix for MALDI Imaging Mass Spectrometry of Phospholipids in Both Ion Modes

TL;DR: The optimized matrix selection and optimized matrix preparation for IMS experiments in both ion modes that maximize PL identification from a single brain tissue section showed improved stability and consistency during both ion mode experiments and successfully identified >100 peaks of PLs determined by parent ion m/z value.

From lipids analysis towards lipidomics, a new challenge for the analytical chemistry of the 21st century. Part II: Analytical lipidomics

TL;DR: The widespread use of soft-ionization mass-spectrometric technology has enabled rapid, sensitive detection of a vast number of individual lipid molecular species from a single biological sample, which enhances development of MS strategies that permit comprehensive analysis of the lipidome.

Alterations in lipid homeostasis of mouse dorsal root ganglia induced by apolipoprotein E deficiency: a shotgun lipidomics study

TL;DR: Four apolipoprotein E‐induced alterations in the lipidome of mouse dorsal root ganglia (DRG) are identified through utilizing the technology of shotgun lipidomics and the potential roles of apoE in lipid transport and homeostasis in a tissue specific manner are suggested, thereby providing insights into the biochemical mechanisms underlying the functions of aposylceramide in the PNS.
