The emerging field of lipidomics
TLDR
Novel analytical approaches — in particular, liquid chromatography and mass spectrometry — for systems-level analysis of lipids and their interacting partners (lipidomics) now make this field a promising area of biomedical research, with a variety of applications in drug and biomarker development.Abstract:
The crucial role of lipids in cell, tissue and organ physiology is demonstrated by a large number of genetic studies and by many human diseases that involve the disruption of lipid metabolic enzymes and pathways. Examples of such diseases include cancer, diabetes, as well as neurodegenerative and infectious diseases. So far, the explosion of information in the fields of genomics and proteomics has not been matched by a corresponding advancement of knowledge in the field of lipids, which is largely due to the complexity of lipids and the lack of powerful tools for their analysis. Novel analytical approaches--in particular, liquid chromatography and mass spectrometry--for systems-level analysis of lipids and their interacting partners (lipidomics) now make this field a promising area of biomedical research, with a variety of applications in drug and biomarker development.read more

Lipid extraction by methyl-tert-butyl ether for high-throughput lipidomics
TL;DR: In this paper, the methyl-tert-butyl ether (MTBE) extraction method was used for shotgun profiling of lipid species, and the results showed that the MTBE protocol delivered similar or better recoveries of species of most major lipid classes compared with the "gold-standard" Folch or Bligh and Dyer recipes.

TL;DR: This work has combined targeted and non-targeted NMR, GC-MS and LC-MS methods with computer-aided literature mining to identify and quantify a comprehensive, if not absolutely complete, set of metabolites commonly detected and quantified (with today's technology) in the human serum metabolome.

Global analysis of the yeast lipidome by quantitative shotgun mass spectrometry
Christer S. Ejsing,Julio L. Sampaio,Vineeth Surendranath,Eva Duchoslav,Kim Ekroos,Robin W. Klemm,Kai Simons,Andrej Shevchenko +7 more
TL;DR: Comparative lipidomics demonstrated that growth temperature and defects in lipid biosynthesis induce ripple effects throughout the molecular composition of the yeast lipidome, establishing shotgun lipidomics as a powerful platform for complementing biochemical studies and other systems-level approaches.

Antagonists of myo-inositol 3,4,5,6-tetrakisphosphate allow repeated epithelial chloride secretion
TL;DR: A series of membrane-permeant Ins(3,4,5,6)P(4) derivatives, carrying alkyl substituents on the hydroxyl groups and screened them for effects on Cl(-) secretion in a human colonic epithelial cell line, T(84), support the hypothesis that Ins(2+) activated Cl(+) secretion stereospecifically inhibits Ca(2%) activated Cl (-) secretion and suggest that antagonists of Ins( 3,4
