Journal ArticleDOI
Degradation of fibrillar collagen in a human melanoma xenograft improves the efficacy of an oncolytic herpes simplex virus vector.
Trevor D. McKee,Paola Grandi,Wilson Mok,Wilson Mok,George Alexandrakis,Numpon Insin,John P. Zimmer,Moungi G. Bawendi,Yves Boucher,Xandra O. Breakefield,Rakesh K. Jain +10 more
TL;DR: Fibrillar collagen can be a formidable barrier to viral distribution and matrix-modifying treatments can significantly enhance the therapeutic response, resulting in an extended range of infected tumor cells and improved virus propagation, ultimately leading to enhanced therapeutic outcome.
Journal ArticleDOI
Fluorescent Nanorods and Nanospheres for Real‐Time In Vivo Probing of Nanoparticle Shape‐Dependent Tumor Penetration
Vikash P. Chauhan,Zoran B. Popović,Ou Chen,Jian Cui,Dai Fukumura,Moungi G. Bawendi,Rakesh K. Jain +6 more
TL;DR: The tumor microenvironment limits the uniform penetration of nanotherapeutics by slowing or halting their transport through hydrodynamic and steric hindrance, and overcoming these physiological barriers in tumors is an outstanding challenge for nanomedicine.
Journal ArticleDOI
PF-06463922, an ALK/ROS1 Inhibitor, Overcomes Resistance to First and Second Generation ALK Inhibitors in Preclinical Models.
Helen Y. Zou,Luc Friboulet,David P. Kodack,Lars D. Engstrom,Qiuhua Li,Melissa West,Ruth W. Tang,Hui Wang,Konstantinos Tsaparikos,Jinwei Wang,Sergei Timofeevski,Ryohei Katayama,Dac M. Dinh,Hieu Lam,Justine L. Lam,Shinji Yamazaki,Wenyue Hu,Bhushankumar Patel,Divya Bezwada,Rosa L. Frias,Eugene Lifshits,Sidra Mahmood,Justin F. Gainor,Timothy Affolter,Patrick B. Lappin,Hovhannes J. Gukasyan,Nathan V. Lee,Shibing Deng,Rakesh K. Jain,Ted William Johnson,Alice T. Shaw,Valeria Fantin,Tod Smeal +32 more
TL;DR: The results suggest that PF-06463922 will be highly effective for the treatment of patients with ALK-driven lung cancers, including those who relapsed on clinically available ALK inhibitors because of secondary ALK kinase domain mutations and/or brain metastases.
Journal ArticleDOI
Transport of fluid and macromolecules in tumors. III. Role of binding and metabolism.
TL;DR: The main conclusion is that an antibody with the highest possible binding affinity should be used except when: there are significant necrotic regions; the diffusive vascular permeability is very small; and a uniform concentration is required on a microscopic scale.
Journal Article
Augmentation of transvascular transport of macromolecules and nanoparticles in tumors using vascular endothelial growth factor.
Wayne L. Monsky,Dai Fukumura,Takeshi Gohongi,Marek Ancukiewcz,Herbert A. Weich,Vladimir P. Torchilin,Fan Yuan,Rakesh K. Jain +7 more
TL;DR: The findings suggest that exogenous VEGF may be useful for augmenting the transvascular delivery of larger antineoplastic agents such as gene targeting vectors and encapsulated drug carriers into tumors.
