TL;DR: In vitro studies with AChE and Na+K+ATPase enzymes indicated that enzymatic activity of AChB was inhibited competitively by fractioned venom in gastrocnemius muscle, and increase in ACP, LDH and GPT enzyme activity was observed after scorpion envenomation.
Abstract: The toxic effects of Asian black scorpion Heterometrus fastigiousus (Family, Scorpionidae) venom were determined in albino mice (NIH strain). Venom was isolated and fractioned by Sepharose CL-6B column chromatography. The toxicity of fractioned venom was determined in albino mice by subcutaneous envenomation. The LD(50) of venom was found to be 15 mg kg(-1) body weight and range of molecular weight of venom proteins responsible for toxicity was found from 9.5-63 kDs. The effects of fractioned venom on different biochemical and enzymatic parameters in blood serum and gastrocnemius muscle tissue of albino mice were determined after experimental envenomation. An increase in serum levels of glucose, free amino acids, uric acid, pyruvic acid and total protein was observed while a decrease in the cholesterol level in serum was observed after 4 h of envenomation. Increase in alkaline phosphatase (ALP), acid phosphatase (ACP), lactic dehydrogenase (LDH) and glutamate-pyruvate transaminase (GPT) enzyme activity in serum was observed. Glycogen content in liver, atria, ventricle, rectus abdominus and gastrocnemius muscle was decreased after experimental envenomation. Activity of ALP, ACP, LDH, GPT, AChE and Na+K+ATPase enzymes in gastrocnemius muscle tissue of envenomed albino mice was studied. Inhibition in ALP, AChE and Na+K+ATPase enzyme activity and increase in ACP, LDH and GPT enzyme activity was observed in gastrocnemius muscle after scorpion envenomation. In vitro studies with AChE and Na+K+ATPase enzymes indicated that enzymatic activity of AChE was inhibited competitively by fractioned venom in gastrocnemius muscle.

TL;DR: In this article, a different approach for the synthesis of N-substituted 2-carboxy-4-quinolones using direct reductive amination followed by LiHMDS-induced cyclocondensation has been developed.
Abstract: A different approach for the synthesis of N-substituted 2-carboxy-4-quinolones using direct reductive amination followed by LiHMDS-induced cyclocondensation has been developed. A range of analogues of the title compounds with broad substrate scope were obtained in moderate yields and good regioselectivity.

TL;DR: In this article, structure optimization and frequency calculation of six nitro derivatives of 1,3,5-triazepine were performed using a MP2(FULL)/6-311G(d,p) method.
Abstract: Structure optimization and frequency calculation of six nitro derivatives of 1,3,5-triazepine were performed using a MP2(FULL)/6-311G(d,p) method. In order to obtain reliable energy data, single-point energy and subsequently thermodynamic properties of the species considered were calculated at a fairly high level of theory, CCSD(T)/6-311G(d,p). Solid-phase heats of formation and crystal density were determined using an electrostatic potential (ESP) method utilizing wave function analysis-surface analysis suite (WFA-SAS) code. The result shows that all nitro derivatives possess high positive heats of formation that increase with an increase in the number of nitro groups attached to the ring moiety. The crystal density was found to be in the range of 1.67–1.90 g/cm3. Detonation properties of the compounds were estimated using the Kamlet-Jacobs equation. The results showed that detonation velocity (D) and detonation pressure (P) increased with an increase in the number of nitro groups attached at the ring mo...

TL;DR: It was observed that the molluscicidal activity of crotocaudin against L. acuminata is time- as well as dose-dependent and there was a significant negative correlation among LC50 values and exposure periods.
Abstract: The compound crotocaudin extracted from the stem bark of the medicinal plant Croton tiglium Linn. was administered for 24 h or 96 h to the freshwater vector snail Lymnaea (Radix) acuminata Lamarck in order to test its toxicity. L. acuminata is the intermediate host of Fasciola hepatica and Fasciola gigantica which cause immense harm to man and his domestic animals. It was observed that the molluscicidal activity of crotocaudin against L. acuminata is time- as well as dose-dependent. There was a significant negative correlation among LC50 values and exposure periods, i.e. increasing the exposure time, the LC50 value of crotocaudin decreased from 5.37 microM (24 h) > 2.08 microM (48 h) > 1.36 microM (72 h) to 1.01 microM (96 h), respectively, against L. acuminata. The toxicological experiments to proof for environmental toxicity, if any, have also been carried out on the non-target freshwater fish Channa punctatus (Bloch) [Channidae (Ophicephalidae)], which shares the habitat with L. acuminata. The sublethal doses of crotocaudin (40% and 80% of LC50) administered over 24 h caused significant changes in the carbohydrate and nitrogenous metabolisms in nervous, hepatopancreas, and ovotestis tissues of Lymnaea acuminata. Channa punctatus was also exposed to sublethal doses of crotocaudin (40% and 80% of 24-h LC50 of L. acuminata) for 96 h which showed significant alterations in the metabolism in muscle, liver, and gonad tissues. After withdrawal of crotocaudin the snail tissues recovered in part after 7 days and the fish tissues completely.
