TL;DR: In this article, two different forms of elastin were characterized for gene expression by quantitative real-time polymerase chain reaction (qPCR) and for protein expression by immunohistochemistry of ex vivo and in vitro reconstituted skin.
Abstract: INTRODUCTION Photoaging is the process by which ultraviolet rays gradually induce clinical and histological changes in the skin through the production and organization of biological molecules, such as elastin, which is critical to skin strength and elasticity. After exposure to radiation, elastin may undergo alternative mRNA splicing, resulting in modified proteins that contribute to the formation of aging characteristics, such as solar elastosis. The present work aimed to study two different forms of elastin under these conditions: normal elastin and elastin that had been altered in exon 26A. METHODS These different forms of elastin were characterized for gene expression by quantitative real-time polymerase chain reaction (qPCR) and for protein expression by immunohistochemistry of ex vivo skins (from photoexposed and non-photoexposed areas) and in vitro reconstituted skin. In addition, up- and downstream molecules in the elastin signaling cascade were evaluated. RESULTS As a result, a significant increase in the gene expression of elastin 26A was observed in both ex vivo photoexposed skin tissues and the in vitro photoexposed reconstituted skins. Additionally, significant increases in the gene expression levels of matrix metalloproteinase-12 (MMP12) and lysyl oxidase (LOX) were observed in the ex vivo skin model. The evaluation of protein expression levels of some photoaging markers on the reconstituted skin revealed increased tropoelastin and fibrillin-1 expression after photoexposure. CONCLUSION This work contributes to a better understanding of the biological mechanisms involved in photoaging, making it possible to obtain new strategies for the development of dermocosmetic active ingredients to prevent and treat skin aging.

TL;DR: Teeth subjected to irrigation with heated sodium hypochlorite showed decreased resistance to crown fracture, indicating contact of the irrigating solution with the root canal walls potentially can cause their weakness, determining crown fracture.
Abstract: Aim This study aimed to evaluate the fracture resistance of tooth crowns endodontically irrigated using different protocols. Materials and methods A total of 76 bovine incisors were divided into four groups (n = 19): irrigation with distilled water (control; CON); conventional irrigation with positive apical pressure (PAP); passive ultrasonic irrigation using continuous flushing (PUI); and irrigation with PAP and heated sodium hypo-chlorite solution (PHS). The force required to fracture the crown was measured on a universal testing machine at an angle of 45°. Results The CON group had higher fracture resistance (351.71 ± 58.66 N) than the PAP and PUI groups (140.96 ± 37.26 N; 167.49 ± 40.08 N respectively). The PHS group had the lowest fracture resistance value (115.15 ± 41.07 N). Irrigation protocols had a significant effect on crown resistance (p Conclusion Teeth subjected to irrigation with heated sodium hypochlorite showed decreased resistance to crown fracture. Clinical significance The contact of the irrigating solution with the root canal walls potentially can cause their weakness, determining crown fracture.

TL;DR: In this paper, the authors analyzed 765 articles published between 2001 and 2006, in the proceedings of EnAnpad's and 3Es in order to evaluate quantitatively the research design mostly used throughout the period.
Abstract: The documentary analysis of publications permits to identify scientific production standards associated with characteristics and tendencies expressed in a certain area of knowledge. Considering the area of Strategy in Organizations and its thematic axes, this study analyzed 765 articles published between 2001 and 2006, in the proceedings of EnAnpad’s and 3Es in order to evaluate quantitatively the research design mostly used throughout the period. Moreover, tendencies expressed in terms of stratification standards and cooperation in the area were also investigated. The data were treated in a descriptive-quantitative way, through statistic procedures and multivariate technique of analysis of multiple correspondence. The findings indicated a predominance of a scientific production focused on empirical articles, mainly case studies and surveys. Additionally, the results revealed the growth of the area concerning the number of articles, authors and institutions, accompanied by greater cooperation in scientific production and changes regarding the regional and institutional stratification standards.

TL;DR: In this paper, the influence of land use parameters defined by local legislation, at the Structural Axes in Curitiba (PR), which are characterized by strong stimulus both to high urban density and vehicular traffic, is assessed.
Abstract: The increase in the number of vehicles causes deterioration of air quality and decline of population health and well-being. The urban grid of buildings influences air flow, hampering the dispersion of vehicular emissions. The present study aims to assess the influence of land use parameters defined by local legislation, at the Structural Axes in Curitiba (PR), which are characterized by strong stimulus both to high urban density and vehicular traffic. Variations in building setback front line and height were considered, specifically on Avenida Sete de Setembro, in order to investigate their influence on pollutant concentration. The conclusion includes suggestions for modifications in the current city by-laws of land use, recommending that building height be reduced on the side opposite to prevailing winds, and increasing, in yet not consolidated stretches, the frontal setback of mezzanines.
