Abstract: This work aimed at evaluating the influence of organic solvents and stationary phases in the extraction with glass beads and chromatographic purification of carotenoids, especially torularhodin, from Sporobolomyces ruberrimus. The combinations of acetone:hexane (1:1 v/v) and acetone:ethyl ether (1:1 v/v) yielded 171.74 and 172.19 μg of total carotenoids.g of cells-1, respectively. The first blend resulted in the highest percent of cell lysis of 57.4%. Among different proportions of acetone:hexane, the 9:1 v/v mixture showed a significant difference (p < 0.05), resulting in a recovery of total carotenoids of 221.88 μg.g of cells−1. The purification of carotenoids was made by preparative chromatography and the yield of the silica-containing stationary phase was higher (24 μg torularhodin.g cells−1). The analyses of the purified fractions in thin layer chromatography and high performance liquid chromatography indicated that the purification of carotenoids, especially of torularhodin, was successfully performed. The combination of polar (acetone) and non-polar solvents (hexane) and the use of silica as stationary phase was efficient to recover and purify torularhodin from the intracellular pigments of Sporobolomyces ruberrimus.

Abstract: O xilitol tem se mostrado um produto de grande valor comercial para a industria alimenticia, farmaceutica e odontologica, em virtude de suas caracteristicas beneficas, como sua docura e a prevencao de caries, simultaneamente a que e indicado para obesos e tolerado por diabeticos. O xilitol pode ser obtido pelas vias quimicas e biotecnologicas. Segundo a literatura, a producao quimica do xilitol envolve um processo dispendioso, pouco produtivo, em resposta ao processo de purificacao. A via biotecnologica, por sua vez, tem-se tornado uma excelente alternativa para sua producao, empregando-se processos fermentativos realizados por microrganismos. Neste trabalho foi estudada a utilizacao da via biotecnologica, utilizando-se a levedura Candida guilliermondii e bagaco de cana-de-acucar como substrato, um residuo agroindustrial de baixo custo empregado para fermentacao da xilose. Realizaram-se ensaios em duplicata, onde foi testada a capacidade fermentativa de tres cepas de C. guilliermondii nos tempos de 0, 24, 48, 72 e 96 horas. Inicialmente, foi realizada hidrolise acida para conversao da xilana em xilose. Utilizou-se, em seguida, carvao ativado para purificacao do hidrolisado e suplementacao do meio de fermentacao com sulfato de amonio e farelo de arroz. O processo fermentativo ocorreu em misturador shaker a 150rpm e temperatura de 30 oC. O estudo teve como objetivo estimar por meio do crescimento das leveduras e o consumo de xilose os melhores parâmetros para producao do xilitol. Entre as cepas utilizadas a de melhor resultado foi a LBP018, onde se obteve a maior quantidade de biomassa e consumo de xilose. Palavras-chave: Xilitol. Fermentacao. Processo biotecnologico.

Abstract: The present study evaluated polymorphisms in RANK, RANKL and OPG-encoding genes to assess whether they are associated with mucositis and peri-implantitis in a population from the Brazilian Amazon region. One hundred and fourteen patients with dental implants were included in the study. After clinical and radiographic examination, the sample was categorized into 4 groups, according to the peri-implant status: Healthy (n=71), Mucositis (n=30), Peri-implantitis (n=13) and Diseased (Mucositis + Peri-implantitis, n=43). Genomic DNA was extracted from buccal cells from saliva, and the genetic polymorphism in osteoprotegerin (OPG), Kappa nuclear factor activator receptor (RANKL) and nuclear kappa factor activator receptor (RANK) were genotyped by the real time PCR. Univariate and multivariate statistical analyses were performed to compare clinical variables among groups and to evaluate genotypes and alleles distributions and the established alpha was 5%. Age, peri-implant biotype, diabetes and presence of peri-implant biofilm were associated with mucositis (p 0.05). The studied genetic polymorphism in RANK, RANKL and OPG were not associated with mucositis and peri-implantitis in a Brazilian population from the Amazon region.

Abstract: This article aims to devise theoretical methodological principles for the organization study. The concept of social practice is adopted as less meaningful unit for analysis of the organization, comprising it from the Theory of Socio-Historical and Cultural Activity, relying on the ideas of Vygotsky, Leontiev, Engestromand Clot. It is argued in this paper the concept of organization from its dialectical and historical events, in which the individual is considered an instance of totality, from a theoretical course in socio - historical categories of: activity systems situated, sensed and meant, learning and human development. The future shows a way of doing science that surpasses the positivist conception of method to result in a conception of method and result. From these theoretical categories and the report of three researches, we can deduce a set of theoretical methodological principles of socio- historical nature that contribute to the study of the organization, as it is broken with the individual dichotomy versus group versus organization, with the abstract and historical conception of the organizational phenomenon, and with the behaviorist conception of prevailing man in the organizational studies. At last, we can deduce categories that learn subjective and intersubjective dimensions of an organization reality in constant movement.
