TLDR
FGF signaling also appears to play a role in tumor growth and angiogenesis, and autocrine FGF signaling may be particularly important in the progression of steroid hormone-dependent cancers to a hormone-independent state.
Abstract
Fibroblast growth factors (FGFs) are small polypeptide growth factors, all of whom share in common certain structural characteristics, and most of whom bind heparin avidly. Many FGFs contain signal peptides for secretion and are secreted into the extracellular environment, where theycan bind to the heparan-like glycosaminoglycans (HLGAGs) of the extracellular matrix (ECM). From this reservoir, FGFs mayact directlyon target cells, or theycan be released through digestion of the ECM or the activityof a carrier protein, a secreted FGF binding protein. FGFs bind specific receptor tyrosine kinases in the context of HLGAGs and this binding induces receptor dimerization and activation, ultimatelyresulting in the activation of various signal transduction cascades. Some FGFs are potent angiogenic factors and most playimportant roles in embry onic development and wound healing. FGF signaling also appears to playa role in tumor growth and angiogenesis, and autocrine FGF signaling maybe particularlyimportant in the progression of steroid hormone-dependent cancers to a hormone-independent state.

Specific binding of basic fibroblast growth factor to basement membrane-like structures and to purified heparan sulfate proteoglycan of the EHS tumor.

TL;DR: Investigation of the binding of iodinated basic fibroblast growth factor to low‐density heparan sulfate proteoglycan purified from the Engelbreth Holm Swarm sarcoma found that it could be strongly inhibited by heparin and was displaced by an excess of unlabeled factor and completely suppressed after heparitinase and hepar inase treatments.

Profound ligand-independent kinase activation of fibroblast growth factor receptor 3 by the activation loop mutation responsible for a lethal skeletal dysplasia, thanatophoric dysplasia type II.

TL;DR: The data suggest that the molecular effect of the TDII activation loop mutation is to mimic the conformational changes that activate the tyrosine kinase domain, which are normally initiated by ligand binding and autophosphorylation.

Structure and Expression of a Novel Member, FGF-16, of the Fibroblast Growth Factor Family ☆

TL;DR: In rat embryos, FGF-16 mRNA was predominantly expressed in the brown adipose tissue, however, the expression decreased greatly after birth, indicating that F GF-16 in embryos might play a role in development of the brown fat tissue.

The int-2 proto-oncogene is responsible for induction of the inner ear

TL;DR: Direct evidence is provided by showing that the formation of the otic vesicle is inhibited by antisense oligonucleotides targeted to the secreted form of int-2, and by antibodies againstint-2 oncoproteins, and basic FGF (bFGF) can mimic the inductive signal in the absence of the rhombencephalon.
