Hypoxia inducible factor-alpha binding and ubiquitylation by the von Hippel-Lindau tumor suppressor protein.

TL;DR: It is shown that extracts from VHL-deficient renal carcinoma cells have a defect in HIF-α ubiquitylation activity which is complemented by exogenous pVHL, and this defect was specific for Hif-α among a range of substrates tested.

Hypoxia-inducible factor-1 (HIF-1) promotes its degradation by induction of HIF-α-prolyl-4-hydroxylases

TL;DR: A direct, negative regulatory mechanism is proposed, which limits accumulation of HIF-1alpha in Hypoxia and leads to accelerated degradation on reoxygenation after long-term hypoxia, which demonstrates functional relevance of the present results.
